139 Improvement of immunological tests for linear IgA bullous dermatosis

Linear IgA bullous dermatosis (LABD) is characterized by linear binding of at the epidermal basement membrane zone. The major autoantigens for LABD are 120 kDa LAD-1 and 97 LABD97, which shedding products BP180. In general, autoantibodies to them detected immunoblotting using cell extract or culture medium keratinocyte lines, such as HaCaT cells, but they not highly sensitive. Therefore, we sought establish more sensitive methods detecting antibodies in LABD. First, checked expression these proteins lines; Ker-CT, HaCaT, DJM-1 primary normal human keratinocytes. Immunoblotting analyses media from cells showed that Ker-CT expressed highest levels BP180, LAD-1, LABD97. We compared sensitivities with 55 sera patients immunoblotting; were 31.5% 38.9%, respectively. LABD97 cover a part 16thnon-collagenous domain (NC16A), contains epitope To evaluate reactivity with/without NC16A, produced four recombinant (RPs) corresponding to; NC16A (490-1421AA), (515-1421AA), without (515-1209AA) mammalian cells. investigated RPs-based enzyme-linked immunosorbent assay (ELISA). Interestingly, some patients’ unique between ELISAs. Among ELISAs, 80% ELISA 490-1421AA RP was sensitivity. Using this ELISA, found autoantibody-titers decreased remission stage 5 6 cases LABD, mucous pemphigoid, IgA/IgG dermatosis. BP180 high-sensitive system its semiquantitative analysis useful clinical decision making.
